I think there are loads of things that are still to be discovered. Often the most important ones are the ones we haven’t yet considered. In physics specifically, I think understanding dark matter will be a huge discovery as it is something that we know almost nothing about and could lead to many other things as well.

Important to what? That makes a big difference to the answer here- it’s almost impossible to guess at the complete most important discovery that hasn’t been made!
To step somewhat into the realm of sci-fi for a while; dark energy seems to have some anti-gravity like behaviour- it is speeding up the expansion of the universe. If we could ever harness it (that’s a big if), could we maybe use it for propulsion of spacecraft? That would be a big discovery!

Honestly, in my view the most important thing we need to discover is a way of making people (countries, nations) on the global scale work together rather fight each other. What we find as a challenge today would be just an easy excersise for the unified effort of the whole humankind. Idealistic, but not unreal!
