I do diagnostic tests for male fertility. I look at how sperm swim, how many there are and their shapes. Sometimes I need to test the pH and check if there are any anti-sperm antibodies.
Many different types but all involving treating liver cells. I can look at changes in response by the cells to different treatments including different doses of metformin. Responses I look at include changes in proteins (are they active or not, is there more or less of them – I use a biochemical technique called Western blotting to do this), the amount of glucose produced, changes in cDNA (complimentary DNA), the changes ATP (the energy currency of the cell), movement of mitochondria (the batteries of a cell where ATP is produced) and other cell structures by imaging movies using a microscope and looking at metformin uptake by cells by using radioactively labelled metformin. All these different types of experiment allow me to get a better idea of what is happening in liver cells (the main site of action of metformin).
